5. When you are ready to execute the concentration method, make sure that you have a blank in the

sampling region but that the series of solutions with different known concentrations are at hand, as you

will receive prompts to insert these samples into the cuvette holder.

6. Now select

Method | Execute

from the menu. A dialog box containing method files opens. Select the

correct concentration method and click

7. As the Palm-SPEC reads the method steps and executes each step in your

concentration method, you will notice the light source turn off (as it

executes method step

3: Store reference and dark spectra

) and the view

switches to Absorbance mode (as it executes method step 4:

View data in

absorbance mode

). You then receive a

Calibration Point

message

prompt similar to the one at right. Make sure that you place the solution

with the correct concentration value into the sample chamber of the Palm-SPEC. Then click the

button in the dialog box.

8. After the Palm-SPEC scans the first sample, you receive a

Calibration Point Stored

message verifying that the sample was

scanned and stored. Click

9. You will continue to receive

Calibration Point

message prompts

and

Calibration Point Stored

confirmation messages for each

sample you stored in the concentration method.

10. Once all of your samples with solution of known concentrations have

been scanned, the Palm-SPEC performs the regression. If you inserted

the method step

6: Display regression results

, you receive a

Calibration Complete

message with the intercept, first coefficient,

second coefficient and R

values. The R

value should be very close to

1.0. If it is not, you may want to perform the method again. Click

11. The view mode switches to concentration. You are now ready to insert samples with solutions of

unknown concentrations. When you need to exit out of the concentration mode, select

Method | Exit

from the menu.
